    • The skin and its appendages, including as hair, nails, feathers, and sebaceous and eccrine glands, make up the integumentary system. The expression profile of connexins and pannexins across the integumentary system in mammals, birds, and fish is the subject of this review. We present a picture of the connexin/pannexin network’s intricacy, as well as the functional role of these proteins in maintaining epidermal barrier integrity. The differential regulation and expression of connexins and pannexins during skin renewal, as well as a number of epidermal, hair, and nail abnormalities linked to connexin mutations, highlight the importance of maintaining a proper balance of connexin and pannexin expression for the skin and its appendages, with both channel and non-channel functions playing important roles. Connexin expression changes during hair and feather regeneration reveal the presence of specialized communication compartments. Finally, we address how zebrafish might be used as a model for connexin skin biology, since evidence grows indicating variable connexin expression plays a role in skin patterning and coloration.
